  In order to get higher extraction efficiency,the polysaccharides from Sparassis crispa were extracted using cellulase.A single-factor study was designed to investigate the effects of different factors on the yield of polysarccharides,such as enzymatic addition,enzymatic hydrolysis temperature,enzymatic hydrolysis time,liquid/material ratio and pH value.Based on the results of single-factor assay,several factors were chosen to be further optimized by response surface methodology.The optimal process parameters are shown as following: pH 5.5,extraction temperature of 40 ℃,enzymatic addition of 1.3 %,hydrolysis time of 2.5 hours,and the material/liquid ratio of 42 mL/g.Under these above conditions,the content of polysaccharides could be up to (22.90 %±0.26 %).The influence of different type macropore resins on decolorization rate and polysaccharide remaining rate of crude polysaccharides extracted on specific conditions,the combination of macropore adsorption resins and macropore anion exchanger got the best results(decoloration rate 80.31% and polysaccharide remaining rate 83.31%).Then with DEAE-52 cellulose was applied to purificate polysaccharide by column chromatography method.The yield of neutral polysaccharides and acid polysaccharides were 7%and 0.9%,respectively.
