人肝癌组织线粒体差异蛋白质组学研究

  目的:比较肝癌手术患者肝癌组织与癌旁组织低丰度线粒体蛋白质表达谱的改变,筛查肝癌相关线粒体蛋白质分子标志物并探讨可能机制。方法:应用双向等电聚焦/聚丙烯酰胺凝胶电泳(IEF/SDS-PAGE)建立20例肝癌切除患者肝癌组织与癌旁组织线粒体蛋白质表达图谱,并进行差异分析,应用基质辅助激光解吸离子化串联时间质谱(MALDI-TOF-TOF/MS)对差异蛋白质点进行鉴定,并进行生物信息学分析。对其中三个差异点的表达变化采用Western blot和免疫组化进行验证。结果:相同条件下对线粒体蛋白样品分别进行3次双向电泳,筛选在三次电泳中均存在的差异点进行分析,在线粒体蛋白质图谱中共鉴定出15个差异蛋白,其中9个蛋白在肝癌组织中低表达,6个蛋白在肝癌组织中高表达。同时采用Western blot,实时荧光定量PCR,石蜡组织芯片和免疫组化对Arg-1的表达进行验证,证实Arg-1在肝癌与癌旁组织中的表达差异与双向电泳的结果一致,并通过临床分析,证实Arg-1与肝癌的发生发展密切相关。结论:肝细胞癌变过程中,不仅许多信号通路蛋白(如Ras-Raf-MEK-ERK)发生了改变,而且一些涉及DNA复制和蛋白折叠的相关蛋白也发生了明显的改变,这些新发现的线粒体蛋白不仅可能成为新的肝癌标志物,而且对后续研究肝癌发生发展的机制提供了前期研究基础。
