产β-甘露聚糖酶内生菌的筛选及酶学特性研究

  β-1,4-甘露聚糖酶(β-1,4-D-mannanmann-ohydrolase)是一类能水解含β-1,4-D-甘露糖糖苷键的内切水解酶,属于半纤维素酶类.自1898年Vogl从黑麦草种子内分离出第一株内生真菌以来,植物内生菌作为一种新的微生物资源受到了广泛的关注.本研究采用富集培养的方法从富含甘露聚糖的植物组织中分离出46株内生菌菌株,利用刚果红染色法进行初筛,选出7株产β-甘露聚糖酶的内生细菌.采用摇瓶培养和测定酶活方法,复筛出2株产酶能力较高的菌株CH-3和HD-1,其酶活分别为42 U/mL和54 U/mL.
