Tyrosinase is a copper-containing enzyme,which is widely distributed in animals,plants and microorganisms.As an important biological resource,tyrosinase is useful in a number of applications and has many important physiological functions in organisms.In this paper,tyrosinase was purified and characterized from the acetone powder of potatoes.Firstly,tyrosinase was separated on gel filtration column and Q FF column by FPLC.Then,purified tyrosinase was characterized by SDS-PAGE.The pH,temperature,and kinetic parameters were studied.The highly purified enzyme could oxidize catechol.L-DOPA was the best substrate.
