Damage to spermatozoa during cryopreservation is regarded as a major obstacle to the expansion of sperm storage technology.The aim of this study was to investigate the exact mechanism of human sperm cryopreservation by using proteomics in order to provide evidence for improvement of human sperm cryodamage.With 2D gel electrophoresis and MALDI-TOF analysis, we identified 26 differentially expressed proteins from the 27 most abundant different protein spots in 2D gels of pre-freeze and post-freeze sperm.Four of which were verified by western blot and localized in sperm by immunofluorescence.The specific protein-related sperm functions such as CASA, vitality, acrosome reaction, capacitation, intracellular ATP content, intracellular [Ca2 +] content, and mitochondrial membrane potential were performed to investigate the relationship between cryodamage and altered proteins.Based on these findings, the authors hypothesized that a-Tocopherol in cryopreserved sperm may protect spermatozoa against capacitation-like change which decreases fertilizing ability.
