Hyperhomocysteinemia decreases Hes1/Hes5 expression in neurons of the frontal cortex of ApoE-/-mice

  An elevated plasma homocysteine level (hyperhomocysteinemia,HHcy) has been reported to be associated with neurode-generative and neuropsychiatric diseases.However,little is known about the brain structure and Hes (hairy enhancer of split)gene expression changes associated with the HHcy.In this study,we investigated brain structure and Hes1 and Hes5 expres-sion in frontal cortex of ApoE-/-mice with HHcy induced with a high-methionine diet and elucidated the changes in brain structure and Hes1/Hes5 expression in cortical neurons associated with HHcy.Brain structure changes were observed by optical and transmission electron microscopy,and Hes1 and Hes5 expression was analyzed by immunohistochemistry,immu-nofluorescence,and western blotting.HHcy (23.71 ± 6.30 μmol/L) was successfully induced by feeding a high-methionine diet for 18 weeks in ApoE-/-mice.Abnormal changes in brain structure and cortical neuron morphology were observed in ApoE-/-mice with HHcy.Reduced ribosomes on rough endoplasmic reticulum and enlarged mitochondria appeared in ApoE-/-mice with HHcy.Hes1 and Hes5 expression was obviously decreased in ApoE-/-mice compared with that in wild-type and ApoE-/-control mice.The results indicate that HHcy caused neuronal damage and decreased Hes1/Hes5 expression in neurons of the frontal cortex ofApo E-/-mice,which likely result in nerve injury.
