  Integral membrane proteins (IMPs) play a critical role in regulating a host of cellular processes, including intercellular communication, vesicle trafficking, ion transport, protein translocation/integration, and propagation of signaling cascades.However, Due to their hydrophobic nature, their analysis presents great challenges not only to solubilization of IMPs, but also to separation of IMPs.Herein, a novel proteomic strategy combining the difference of the solvent solubility based prefractionation with nano-LC-ESI/MS/MS system was developed, and applied for high efficient analysis of IMPs from rat hippocampus, by which protein prefractionation by different solvent extraction could decrease the complexity of sample prior to LC-MS analysis, thus allowing a greater number of peptides and proteins to be detected[1-3].
