  Objectives: To explore the influence of toll/interleukin 1 receptordomain containing protein encoded by E.coli (TcpC) on the apoptosis of kidney cells,so as to provide new evidence to clarify the pathogenicity of TcpC producinguropathogenic E.coli.Materials and methods: 5x105 cells of HEK293,a human embryonic kidney cellline cells,were co-cultured,in transwell,with 5x106 cells of TcpC-producing E.colistrain CFT073 or tcpC-knock out CFT073 mutant strain (CFT073AtcpC) for differentperiod of time.The ceils were harvested and used for DAPI staining and analysis ofapoptosis by Annexin V and PI double staining.Mitochondrial membranedepolarization was determined by JC-1 staining.Levels of caspase-3,PI3K,AKT,XIAP,PARP in the treated HEK293 were analyzed by Western blot.To observe therole of AKT in TcpC-induced apoptosis,AKT over-expression HEK293 cells wereestablished by pTV/CMV-AKT transfection.
