  Extracellular matrix (ECM),including collagens,laminin,or fibronectin,plays an important role in peripheral nerve regen-eration.Recently,Schwann cell (SC)-derived ECM with classical biomaterial was used to mimic a neural niche;however,extensive clinical use of SCs is limited due to limited origin,sacrifice of an autologous nerve,and long in vitro culture time.In the present study,we investigated human umbilical cord-derived mesenchymal stem cells (hUCMSCs),which are easily accessible and more proliferative than SCs.A protocol was developed to prepare and characterize hUCMSC-derived ECM.Compared with non-coated dish tissue culture,hUCMSC-derived ECM enhanced SC proliferation,upregulated the gene and protein expression levels of brain-derived neurotrophic factor (BDNF),glial cell derived neurotrophic factor (GDNF)and vascular endothelial growth factor (VEGF) in SCs,and enhanced neurite outgrowth from dorsal root ganglion (DRG) neurons.These findings suggest that hUCMSC-derived ECM promotes peripheral nerve repair and therefore can be used as a basis for a more rational design of tissue-engineered neural niches.
