由于菌株特异的内切酶能够将外源DNA 酶切降解而阻止菌株接受外源DNA,因此限制修饰系统被推测是影响菌株转化的重要制约因素之一。本实验使用枯草芽孢杆菌ATCC 9372 和地衣芽孢杆菌ATCC 14580 的细胞提取液对pKSV7 进行体外限制性分析,然后对其进行体外甲基化处理,保护其不会被菌株的限制性修饰系统降解,并使其成功转化相应的菌株。此方法解决了菌株难以转化的难题,为不同菌株的基因功能研究
