Many stimuli, including growth factor deprivation, ER stress and oncogenesis can activate autophagic machinery.The reports that the cells underwent apoptosis accompanying autophagy are accumulating.Previously, we repored that overexpression of oncogene Ha-rasval1 2 could trigger apoptosis of the NIH3T3 cells under starvation conditions.In this report, we unravel that under normal conditions, overexpression of Ha-rasval12 oncogene induced cell autophagy.Using lac inducible system to control Ha-rasval12 oncogene expression, we demonstrated that Harasval12 overexpression in NIH3T3 cells could only induce the early-stage of apoptosis.However, it did not trigger apoptosis related cell death.Autophagic machinery as well as calcium concentration and ER stress were actively induced by Ha-rasval12overexpression at 48 hr post IPTG induction.In summary, the response of early-stage apoptosis seems essential for Ha-ras overexpression induced autophagy.ER stress and intracellular calcium concentration may be involved in Ha-ras induced autophagy.The exploration of the role of autophagy in Ha-rasva112 related tumorigenesis is undergoing and will be presented.
