Characteristics of enzyme transition state analogues are reviewed for a range ofreactions.Tetrafluoroaluminate is widely employed as a transition state(TS) mimic but itnecessarily imposes octahedral geometry on the assembled complexes whereas phosphoryltransfer involves trigonal bipyramidal,tbp,geometry.Trifluoromagnesate,MgF3-,providesthe near-ideal solution,delivering tbp geometry and correct anionic charge),2 TS analoguesfor a range of kinases,mutases,and phosphatases provide a detailed description of mechanismfor phosphoryl group transfer,supporting the concepts of the anionic shield for ground statestability and of charge balance for their TSs and concerted-associative pathways forbiocatalysis.
