  Physiologically based pharmacokinetic (PBPK) modelling is a simulation tool that has a huge impact in the Lead optimization stage.In PBPK modelling,each tissue is represented as a compartment that has an associated blood flow rate,volume and a tissue partition coefficient.Drug transport between these compartments is driven by real blood flow rates and membrane permeabilities.Thus the model attempts to mimic and mathematically model all the physical and biophysical processes that determine the fate of the drug in the body.The dynamics of the drug within each tissue,as it permeates the plasma membrane,gets eliminated in the tissue or carried forward are represented by differential equations and solved with mathematical software such as MATLAB.PBPK looks at integrating all available compound information in the context of the physiology of the species to predict time profiles of plasma and tissue concentrations.It is aimed at estimating concentrations reaching liver resulting from first pass of an orally administered drug and concentration in tissues.
