Previous studies indicated that lincRNA-ROR play an important role in the maintenance of induced pluripotent stem cells (iPSCs) and embryonic stem cells (ESCs), and regulate tumor carcinogenesis and metastasis.Although increasing studies focus on the role of lincRNA-ROR in BC progression, it remains unknown whether circulating lincRNA-ROR could function as a potential biomarker for BC diagnosis and monitoring.In current study, we firstly carried out a reverse transcription-quantitative polymerase chain reaction (RT-qPCR) to examine the expression levels of lincRNA-ROR in BC tissues and cell lines.We found that lincRNA-ROR expression level was significantly increased in BC tissues compared with adjacent normal tissues (p <0.01) and lincRNA-ROR was also highly expressed in BC cell lines compared with that in immortalized mammary epithelial cells (MECs) MCF10A (all p < 0.001) .Furthermore, we investigate the differentially expressed of lincRNA-ROR in another 94 BC plasma samples and 90 healthy controls by RT-qPCR.The results revealed that the plasma lincRNA-ROR level was significantly upregulated in BC patients than health controls, and plasma lincRNA-ROR levels were significantly associated with estrogen receptor (ER) (p =0.042) and lymph node metastasis (p =0.046).Receiver operating characteristic (ROC) curve was conducted to evaluate the diagnostic values of plasma lincRNA-ROR for BC patients.The area under the ROC curve (AUC) of plasma lincRNA-ROR was 0.844 (sensitivity, 80.0%;specificity, 56.7%;p < 0.0001), which was higher than the values of carcinoembryonic antigen (CEA) and carbohydrate antigen 15-3 (CA15-3).Lastly, we further to explore the tumor monitoring values.The result showed that plasma lincRNA-ROR levels were significantly decreased in postoperative samples compared with preoperative samples (p < 0.0001) and plasma lincRNA-ROR levels moderately correlated with the corresponding tissues levels of the same patients (r2 =0.638, p < 0.0001).In conclusion, our results suggested that the increased expression of lincRNA-ROR in plasma may be a potential biomarker for BC early diagnosis and dynamic monitor.
