Approaches in TCM research using monoclonal antibodies against natural products

  Recently the preparation of monoclonal antibodies (MAbs) against natural products have been increasing.We also started to prepare MAb from 1993 and accumulating many kind of MAb like against ginsenosides, saikosaponin, glyeyrrhizin, berberine, aeonitine, sennosides,forskolin, liquiritin, artemisinin, aristolochic acid and so on.The first approach is eastern blotting newly named using MAb.Natural products containing glycoside in a molecule were developed by TLC and the TLC plate was covered by PVDF or PES membrane and blotted.The membrane was treated with NaIO4, and then with carrier protein like BSA resulting in glycoside-carrier protein conjugate on the membrane.Peroxidase labeled secondary MAb and then substrate were added successively for staining.In the case of natural product without glycoside in a molecule for example aristolochic acid can be conjugated with carrier protein via carboxylic acid resulting in fixing on the membrane and staining.One step isolation of natural product from crude extracts by an affinity column using MAb is the second approach.We succeeded to isolate the target compounds (antigen) by one step purification like forskolin, solasodine glycosides, ginsenoside Rbl, glycyrrhizin and so on.In the case of glycyrrhizin the knockout extract named that is the eluted fraction by washing solvent can be used for further experiments like synergic activity.As the third approach the histochemical investigation is available.The distribution of ginsenoside Rbl in ginseng tissue was confirmed by anti-ginsenoside Rbl MAb.The anti-aristolochic acid MAb can determine the distribution of aristolochic acid in mice kidney tissues.Furthermore, we determined a targeting protein conjugated with aristolochic acid in the human kidney cells by using anti-aristolochic acid MAb.

目的:建立测定牛黄乌精胶囊中胆红素含量的方法.方法:采用HPLC法,色谱柱为ZORBAX SB-C18(4.6×150mm,51μm);流动相:乙腈-1%冰醋酸(95∶5);检测波长:450nm;柱温:30℃;流速:1.0ml/min;进样量:5μ1.结果:胆红素在5μg/ml~50μg/ml浓度范围内有良好线性关系(r=0.9995),平均加样回收率为100.91%,RSD%=1.82%.结论:

车前子(PlantaginisSemen)为常用中药,具有清热利尿通淋、渗湿止泻、明目、祛痰等功效.现代药理学研究表明,车前子具有降血压、降糖、降脂等作用,但其药效物质基础、作用机理不明确.本课题组利用多种代谢综合症动物模型及体外模型,对车前子提取物治疗代谢综合症的药效进行了评价,并对其活性成分进行了筛选.以基因表达谱芯片和RT-PCR技术,对车前子治疗代谢综合症的作用机理进行了探讨.
