Abscisic acid(ABA)is a key endogenous phytohormone in plant development and responses to various stresses.In this study,a transcription factor OsRE1 was identified to regulate methylerythritol phosphate(MEP)pathway and β-carotene accumulation in rice(Oryza sativa L.).Transgenic rice callus over-expressing OsRE1 showed in orange color,indicating that β-carotene biosynthesis pathway may be regulated by OsRE1.Expression profile analysis revealed that OsDXR and OsDXS,encoding two critical enzymes in MEP pathway,were up-regulated,and β-carotene,a substrate in MEP pathway,was accumulated in the OsRE1-overexpression rice plants.Sub-cellular location assay in rice protoplasts indicated that OsRE1 is a nuclear protein.Transcriptional activation assay in rice protoplasts suggested that OsRE1 could activate the expression of OsDXS and OsDXR promoter-driven luciferase.Moreover,7-day old OsRE1-overexpression seedlings showed significantly improved sensitivity to ABA treatment.Based on sequence search,two homologs of OsRE1,named OsRE2 and OsRE3,were identified in rice genome.Over-expression of OsRE2 and OsRE3 also showed β-carotene accumulation.These findings reveal that OsRE1 is a new transcription factor that regulates the MEP pathway which is involved in β-carotene accumulation in rice.
