草鱼葡萄糖激酶基因全长cDNA分子克隆及组织表达研究

  采用RT-PCR和RACE技术,从草鱼肝胰脏中克隆了葡萄糖激酶(GK)基因全长cDNA.该cDNA全长2066bp,含有1个1431bp的开放阅读框,编码476个氨基酸.GK蛋白计算分子量53.7kDa,等电点5.11.该蛋白保守功能位点包括:1个保守的已糖激酶标签序列Leu156-Phe181,2个N连接糖基化位点Asn176和Asn214,1个细胞粘附序列Arg202-Asp204,1个糖胺聚糖附着位点Ser455-Gly458.GK氨基酸序列与其他脊椎动物相似度很高,与翘嘴红鮊、鲤、人和鼠的相似百分比分别为98.1%、96.8%、80.3%和79.8%.采用SYBR实时荧光定量PCR技术对GK基因在大脑、肠系膜脂肪组织、肾脏、白肌和肝胰脏等组织中的表达进行了研究,结果表明:肝胰脏中GK基因的表达量显著高于肠系膜脂肪组织、肾脏和大脑(P<0.05),经β-actin校准后,肝胰脏中GK mRNA表达量分别为肠系膜脂肪组织、肾脏和大脑的31、454和649倍,白肌中未检测到GK mRNA表达.
