  Objective:To investigate the proper beginning exercise time on cerebral ischemia rat,we evaluated peri-infarct region inflammation in acute phase and neurological function,infarct volumes,cell death,apoptosis and autophagy in subacute phase following ischemic stroke.Methods:SD rats were randomly divided into three groups,including sham,control and exercise groups while exercise group and control group were further divided into four subgroups,respectively,i.e.,(1)EX24h,(2)EX48h,(3)EX72h,(4)EX96h subgroups(starting exercise within 24h,48h,72h,96h after ischemic stroke,respectively)and(1)Con24h,(2)Con48h,(3)Con72h,(4)Con96h subgroups(corresponding control subgroups compared with exercise subgroups).Rats in exercise and control groups were subjected to transient middle cerebral artery occlusion(tMCAO).The expressions of interleukin-1α(IL-1α),IL-1β,IL-6,IL-10 and tumor necrosis factor-α(TNF-α)were examined by quantitative real time polymerase chain reaction(qRT-PCR)at 3 days after exercise.The modified neurological severity score(mNSS)was measured at 1 day post stroke,7 and 14 days after exercise.Nissl staining evaluating infarct volume and cell death(or the ratio of intact cells),together with immunofluorescence assessing cell apoptosis and autophagy,was applied at 14 days after exercise.Results:The EX24h subgroup showed no significant difference of mNSS and infarct volumes with Con24h subgroup while EX48h(p < 0.05),EX72h(p < 0.05),EX96h(p < 0.05)subgroups attenuated the mNSS and infarct volumes compared with corresponding control subgroups.No significant difference of mNSS was found among exercise subgroups.However,infarct volume in EX24h subgroup(p < 0.05)increased amid exercise subgroups.In contrast with corresponding control subgroups,down-regulated expressions of IL-1α,IL-1β,TNF-α(P < 0.05),and up-regulated expression of IL-10(P < 0.05)were found in EX48h,EX72h,EX96h subgroups.By contrast,increased expressions of IL-1α,IL-1β,TNF-α were found in EX24h subgroup(P < 0.05),compared with Con24h subgroup.Among exercise subgroups,EX24h subgroup showed increased expressions of IL-1α,IL-1β,TNF-α(P < 0.05)and decreased expression of IL-10(P < 0.05)while EX48h subgroup showed decreased IL-1β expression(P < 0.05)and increased IL-10 expression(P < 0.05).In contrast to corresponding control subgroups,the number of LC3,TUNEL positive cells decreased(P < 0.05)and intact cells expression increased(P < 0.05)in EX48h,EX72h,EX96h subgroups,but no significant difference was found in EX24h subgroup.Compared with EX24h subgroup,the reduction of LC3,TUNEL positive cells and the increase of intact cells were found in EX48h,EX96h subgroups(P < 0.05).Conclusions:Our study indicates that exercise starting at 48 hours post stroke may be proper to tMCAO rats,which may be related with decreased inflammation,cell death,apoptosis and autophagy in peri-infarct region.
