目的:模拟现代人“起居无常、饮食失节”致生物钟紊乱现象,对不节律生活致亚健康大鼠模型进行探索。方法:打破大鼠正常生活节律,给予大鼠长期24h/24h间歇睡眠剥夺联合饮食失节法造成亚健康状态。采用单数日24h起居自由+高糖高脂饲料(过食肥甘)、偶数日24h睡眠剥夺+白菜(单一素食)方法造模。比较正常组与模型组大鼠行为学指标、血液学指标、心电学的差异,并观察模型可逆性。结果:与正常组比较,1行为学:模型组自造模第2周出现耳赤现象、抓力显著降低,自第4周痛阈显著降低,自第5周面温显著升高;停止造模1周后面温恢复正常水平、痛阈、抓力无明显改善。2血常规:自造模第5周模型组白细胞(WBC)水平显著降低,血小板分布宽度(PDW)、大血小板比率(P-LCR)显著升高,停止造模1周后血小板分布宽度(PDW)及大血小板比率(P-LCR)恢复正常,白细胞水平(WBC)未见明显恢复。3血生化:自造模第5周血糖(GLU)、总胆固醇(TC)显著增高,甘油三酯(TG)显著降低;停止造模1周后血糖水平(GLU)及甘油三酯水平(TG)恢复正常,总胆固醇(TC)未见明显恢复。4心电:模型组P波高度明显上升,R波高度、QRS波群宽度显著上升,平均心电轴右偏(QRSa);5环核苷酸系统及细胞因子:模型组大鼠血浆c AMP水平及c AMP/c GMP比值显著升高,IL-2水平显著降低,IL-6水平显著升高。结论:采用长期间歇睡眠剥夺联合饮食失节复合因素造模可令大鼠出现耳赤、易惊、乏力等体虚症状,并使大鼠血糖血脂代谢紊乱、血液学指标及心电波形异常、细胞因子和环核苷酸系统紊乱,与人体亚健康表现较接近,可为补益中药干预亚健康等研究提供新的思路或方法。
OBJECTIVE: To simulate the disorder of the biological clock caused by modern people “living impermanence and eating disorders ”, and to explore the sub-healthy rat model of rhythmless life. Methods: The normal circadian rhythm of rats was broken, and the long-term 24h / 24h intermittent sleep deprivation was used to induce sub-healthy state in rats. The differences of behavior, hematology and electrocardiogram between the normal group and the model group were compared, and the reversibility of the model was observed. Results: Compared with the normal group, behavioral study showed that the model group had ear-red phenomenon in the second week of modeling, the grasping force decreased significantly, the pain threshold decreased significantly from the 4th week and the surface temperature increased significantly from the 5th week, After 1 week, the facial temperature returned to normal level, pain threshold and grasping power had no obvious improvement. (2) Blood routine: The WBC level, PDW and P-LCR in the model group decreased significantly at the fifth week after modeling, and the platelet distribution width (PDW) ) And large platelet ratio (P-LCR) returned to normal, no significant recovery of white blood cell (WBC). Blood biochemistry: The blood glucose (GLU), total cholesterol (TC) increased significantly and triglyceride (TG) decreased significantly in the 5th week after modeling. The level of GLU and triglyceride ) Returned to normal, no significant recovery of total cholesterol (TC). 4 ECG: the model group, P wave height increased significantly, R wave height, QRS complex width increased significantly, the average ECG axis deviation (QRSa); 5 nucleotides and cytokines: model group rats plasma c AMP Level and c AMP / c GMP ratio was significantly increased, IL-2 levels were significantly lower, IL-6 levels were significantly higher. Conclusion: Long-term intermittent sleep deprivation in combination with dietary nociceptive factors can cause symptoms such as ear-aching, easy-to-shock, fatigue and so on. It can also lead to disorder of blood glucose, blood lipid, hematology and ECG abnormalities, Factors and cyclic nucleotide system disorders, and the performance of human sub-health is closer, for the benefit of Chinese medicine intervention sub-health and other research provide new ideas or methods.
