  Objective: To evaluate changes of expression in spinal dorsal horn of rat chronic post-thoracotomy pain (CPTP) model.Methods: 20 adult male Sprague-Dawley rats, weighing 250 ~350g, were randomly divided into two groups (10 for CPTP group and 10 for sham group).All of the rats were measured mechanical threshold the day before surgery.Then from the first day post-surgery, all animals were tested for 30 days for mechanical allodynia using calibrated Von Frey filaments and cold allodynia using acetone applied to the incision site.The results were compared between two groups.The thoracic spinal cords of hyperalgesic rats from CPTP group and rats from sham group(both obtained at 30 day post-surgery) were transversely sectioned, the slices were investigated by KATP immunohistochemistry.The expression densities of KATP in spinal dorsal horn neurons were compared between two groups.Results: From the 5 day post-surgery, allodynia was developed, and was kept to the 30 day post-surgery in 7 of 10 CPTP group animals, but none in sham animals.At the day 30 after surgery, the expression density of KATp in spinal dorsal horn neurons was decreased significantly in CPTP group, compared to the sham group.Conclusion: The rib-retraction model seemed to be a stable and reliable model of chronic post-thoracotomy pain, the expression of KATP channel was decreased in the CPTP group, it suggested KATp channel may play a role in the pathogenesis of chronic postthoracotomy pain.
