目的 探讨 APC基因在大肠肿瘤发生、发展中的作用和意义。方法 采用免疫组化方法检测 5 5例大肠腺癌、33例大肠腺瘤组织中 APC基因蛋白产物的表达水平。结果 APC基因蛋白产物在腺癌组织中呈显著低表达 (2 2 .8% ) ,与大肠腺瘤 (72 .7% )和正常粘膜组织 (86 .7% )对比差别有显著性 (P<0 .0 5 )。绒毛状腺瘤阳性表达率明显低于管状腺瘤和混合型腺瘤。大肠癌组织 APC表达与患者的性别、年龄、肿瘤的部位、大体类型、分化程度、 Dukes分期、肿瘤大小以及淋巴结转移情况无关。结论 APC蛋白与大肠粘膜上皮细胞的成熟有关。 APC蛋白产物的失表达与大肠肿瘤的发生有关。免疫组化方法检测 APC蛋白产物尚不能单独用于大肠肿瘤的早期诊断
Objective To investigate the role and significance of APC gene in the development and progression of colorectal tumors. Methods Immunohistochemistry was used to detect the expression of APC gene protein in 55 cases of colorectal adenocarcinoma and 33 cases of colorectal adenoma. Results The APC gene protein product was significantly lower in adenocarcinoma tissues (22.8%) than in adenomas (72.7%) and normal mucosa tissues (86.7%) (P < 0 .0 5). The positive rate of villous adenoma was significantly lower than that of tubular adenoma and mixed adenoma. The expression of APC in colorectal cancer tissue was not related to the patient’s gender, age, tumor location, general type, differentiation, Dukes stage, tumor size and lymph node metastasis. Conclusion APC protein is related to the maturation of colonic epithelial cells. Loss of expression of APC protein products and the occurrence of colorectal cancer. Immunohistochemical detection of APC protein products can not be used alone for the early diagnosis of colorectal tumors
