目的:研究人胃癌HGC-27细胞及其肿瘤球细胞在裸小鼠皮下产生肿瘤的能力。方法:用DMEM培养液(SSM)培养HGC-27细胞。用含有生长因子的DMEM/F12培养液(SFM)在低吸附6孔培养板中诱导肿瘤球的生长。分别将溶于200μL PBS的1×102、1×103、1×104、1×105、1×106和1×107肿瘤细胞或肿瘤球细胞注射到裸小鼠皮下。常规方法制作组织切片。结果:在SSM培养液中HGC-27细胞呈贴壁、伸展状态生长。在SFM及低吸附的培养条件下观察到了肿瘤球的形成,呈悬浮状态。注射了1×106、1×107肿瘤细胞的裸小鼠产生了移植瘤,注射了1×104、1×105、1×106和1×107肿瘤球细胞的裸小鼠产生了移植瘤。移植瘤组织癌细胞间连接疏松,瘤细胞分化程度低。结论:HGC-27肿瘤球细胞比非肿瘤球HGC-27细胞具有更强的致瘤能力。
Objective: To study the ability of human gastric cancer HGC-27 cells and their tumor cells to produce tumor subcutaneously in nude mice. Methods: HGC-27 cells were cultured in DMEM medium (SSM). Growth of tumor spheres was induced in DMEM / F12 medium containing growth factors (SFM) in low adsorption 6-well plates. 1 x 102, 1 x 103, 1 x 104, 1 x 105, 1 x 106, and 1 x 107 tumor cells or tumor spheres, respectively, dissolved in 200 μL of PBS were injected subcutaneously into nude mice. The conventional method of making tissue sections. Results: HGC-27 cells were adherent and expanded in SSM medium. The formation of tumor spheres was observed under SFM and culture conditions with low adsorption, in suspension. Nude mice injected with 1 × 10 6, 1 × 10 7 tumor cells produced transplanted tumors and nude mice injected with 1 × 10 4, 1 × 10 5, 1 × 10 6, and 1 × 10 7 tumor cells produced transplanted tumors. Transplanted tumor tissue loose connective cells, low degree of tumor cell differentiation. Conclusion: HGC-27 tumor cells have stronger tumorigenicity than non-tumor HGC-27 cells.
