当归多糖及小分子提取物对TGF-β1诱导的HELF表达α-SMA、CTGF的影响

目的:观察当归多糖及小分子提取物对TGF-β1诱导的人胚肺成纤维细胞HELF的增殖及α-SMA、CTGF影响的作用。方法:建立肺纤维化细胞模型,分为空白组、模型组、12.5μg/ml当归多糖组、25μg/ml当归多糖组、50μg/ml当归多糖组、25μg/ml当归小分子提取物组、50μg/ml当归小分子提取物组、100μg/ml当归小分子提取物组。结合形态观察,检测细胞增殖、羟脯氨酸、MMP-9、TIMP-1含量及α-SMA、CTGF蛋白表达的变化。结果:与空白组比较,各组细胞吸光度值、羟脯氨酸、MMP-9、TIMP-1含量、CTGF、α-SMA蛋白表达均明显增高。与模型组比较,25μg/ml当归多糖组、50μg/ml当归小分子提取物组细胞吸光度值、羟脯氨酸含量、CTGF、α-SMA蛋白表达均降低,50μg/ml当归小分子提取物组MMP-9含量下降,25μg/ml当归多糖组、50μg/ml当归小分子提取物组TIMP-1含量均增高。形态学观察,模型组细胞数量增多,体积增大,生长旺盛。25μg/ml当归多糖组、50μg/ml当归小分子提取物组细胞数量较少,体积较小。结论:当归多糖及小分子提取物能抑制TGF-β1诱导的HELF增殖,减少羟脯氨酸的合成,其作用可能与下调α-SMA、CTGF表达,调节维持MMP-9/TIMP-1之间的平衡有关。 OBJECTIVE: To observe the effects of Angelica sinensis polysaccharide and small molecule extract on the proliferation of HELF induced by TGF-β1 and the effects of α-SMA and CTGF on human embryonic lung fibroblasts. Methods: Pulmonary fibrosis model was established and divided into blank group, model group, 12.5μg / ml Angelica polysaccharide group, 25μg / ml Angelica polysaccharide group, 50μg / ml Angelica sinensis polysaccharide group, 25μg / ml Angelica sinensis small molecule extract group, 50μg / ml Angelica small molecule extract group, 100μg / ml Angelica small molecule extract group. Combined with morphological observation, the changes of cell proliferation, hydroxyproline, MMP-9, TIMP-1 and the expression of α-SMA and CTGF were detected. Results: Compared with the blank group, the cell absorbance, hydroxyproline, MMP-9 and TIMP-1, CTGF and α-SMA protein expression in each group were significantly increased. Compared with the model group, the cell absorbance value, hydroxyproline content, CTGF and α-SMA protein expression in the 25μg / ml Angelica Sinensis polysaccharide group and the 50μg / ml Angelica Sinensis small molecule extract group were decreased, while 50μg / ml Angelica Sinensis small molecule extract group MMP-9 content decreased, 25μg / ml Angelica polysaccharide group, 50μg / ml Angelica small molecule extract group increased the content of TIMP-1. Morphological observation, the model group increased the number of cells, the volume increased, vigorous growth. 25μg / ml Angelica polysaccharide group, 50μg / ml Angelica small molecule extract group of small number of cells, the smaller the volume.
