目的观察原花青素(Procyanidins,PC)对高糖诱导下晶状体上皮细胞中Bcl-2与Bax表达的影响,探讨原花青素影响晶状体上皮细胞凋亡的机制。方法取发育正常的兔眼晶状体进行体外培养,随机分为空白对照组(A)、高糖处理组(B)、原花青素给药组(C)。通过HE染色观察晶状体形态的变化,应用免疫组化技术检测培养48 h,72 h,96 h晶状体上皮细胞中Bcl-2与Bax的表达情况并进行定量分析。结果高糖使晶状体纤维断裂,上皮细胞结构不完整,原花青素能够减轻病变。随着培养时间的延长,各组Bcl-2的表达逐渐减弱,其中以B组最为明显,C组减弱程度轻于B组。各组Bax表达逐渐增强,以B组最为明显,C组Bax表达增强程度轻于B组。结论原花青素能够下调高糖条件下兔晶状体上皮细胞中Bax的表达,上调Bcl-2的表达,可能通过调控Bcl-2与Bax影响细胞凋亡。
Objective To investigate the effect of procyanidins (PC) on the expression of Bcl-2 and Bax in lens epithelial cells induced by high glucose and to explore the mechanism of procyanidins on the apoptosis of lens epithelial cells. Methods Normal eyes of rabbit lens were cultured in vitro and randomly divided into blank control group (A), high glucose treatment group (B) and proanthocyanidin treatment group (C). The morphological changes of lens were observed by HE staining. The expression of Bcl-2 and Bax in lens epithelial cells cultured for 48 h, 72 h, 96 h were detected by immunohistochemistry and quantified. The results of high sugar to break the lens fibers, epithelial cell structure is not complete, proanthocyanidins can reduce the disease. With the prolongation of culture time, the expression of Bcl-2 gradually decreased in each group, especially in group B, and in group C weaker than that in group B. Bax expression gradually increased in each group, the most obvious in group B, C group Bax expression was lighter than the B group. Conclusions Proanthocyanidins can down-regulate the expression of Bax and up-regulate the expression of Bcl-2 in rabbit lens epithelial cells under high glucose conditions, which may affect the apoptosis through regulating Bcl-2 and Bax.
