目的:探讨滑膜炎颗粒对Ⅱ型胶原诱导的关节炎(CIA)模型改善作用机制。方法:牛Ⅱ型胶原乳剂大鼠尾根部皮下注射制备CIA模型,造模成功后按随机数字表法分为对照组(Ⅰ)、模型组(Ⅱ)、不完全弗氏佐剂组(Ⅲ)、雷公藤多苷片组(Ⅳ)、醋酸泼尼松龙组(Ⅴ)、滑膜炎颗粒大剂量组(Ⅵ)、中剂量组(Ⅶ)、小剂量组(Ⅷ),每组各10~11只大鼠。免疫组化方法观察大鼠左膝关节滑膜组织IL-1、TNF-α、Erk、Jnk、Ras、Raf的表达,图像分析软件计算阳性面积比。结果:各造模组大鼠膝关节滑膜组织IL-1、Erk、Jnk、Ras、Raf表达量均较对照组增高;各给药组Erk表达量均较模型组降低,其中Ⅲ、Ⅳ、Ⅴ、Ⅵ组Erk表达量与模型组比较差异有统计学意义。结论:IL-1、Erk、Jnk、Ras表达升高,参与CIA模型大鼠滑膜组织病理损伤过程;滑膜炎颗粒改善关节损伤症状可能与降低Erk的表达相关,而与IL-1、TNF、Jnk、Ras、Raf不相关。
Objective: To investigate the mechanism of the effect of synovitis granule on the collagen Ⅱ-induced arthritis (CIA) model. Methods: CIA model was established by subcutaneous injection of bovine type Ⅱ collagen emulsion into the caudal root of rats. The rats were divided into control group (Ⅰ), model group (Ⅱ), incomplete Freund ’s adjuvant group (Ⅲ) (Ⅳ), prednisolone acetate group (Ⅴ), synovitis granule high dose group (Ⅵ), middle dose group (Ⅶ), low dose group (Ⅷ) ~ 11 rats. Immunohistochemistry was used to observe the expression of IL-1, TNF-α, Erk, Jnk, Ras and Raf in synovium of left knee joint. The ratio of positive area was calculated by image analysis software. Results: The expression of IL-1, Erk, Jnk, Ras and Raf in synovium of knee joint synovium of rats in each model group was higher than that in control group. The expression of Erk in each group was lower than that in model group, The expression of Erk in group V and group V was significantly different from that in model group. Conclusions: The expression of IL-1, Erk, Jnk and Ras is increased, which is involved in the pathological process of synovial tissue in CIA rats. The synovial tissue injury may be related to the decrease of Erk expression, , Jnk, Ras, Raf are not relevant.
