目的提高毛细管电泳法的灵敏度,建立测定中成药中微量士的宁的高效毛细管电泳法。方法采用高效毛细管电泳场放大富集法,运行缓冲液为70 mmol·mL-1硼砂-乙腈(83∶17,pH5.5),10 kV电动进样,进样时间16 s,分离电压25 kV,分离温度25℃,检测波长205 nm。结果士的宁0.2~16.0μg·mL-1与峰面积的线性关系良好(r=0.9987),检测限为0.022μg·mL-1。结论所用方法准确、简便,有效提高了毛细管电泳法的灵敏度。
Objective To improve the sensitivity of capillary electrophoresis and to establish a high performance capillary electrophoresis method for the determination of trace amount of strychnine in proprietary Chinese medicines. Methods High performance capillary electrophoresis (FCE) enrichment was used. The running buffer was 70 mmol · mL -1 borax-acetonitrile (83:17, pH 5.5), 10 kV electrokinetic injection, injection time was 16 s, separation voltage was 25 kV , The separation temperature of 25 ℃, detection wavelength of 205 nm. Results Strychnine 0.2 ~ 16.0μg · mL-1 and the peak area of a good linear relationship (r = 0.9987), the detection limit was 0.022μg · mL-1. Conclusion The method used is accurate, simple and effective, which improves the sensitivity of capillary electrophoresis.
