目的:观察酪丝亮肽对人肝癌BEL-7402的抑制作用,并分析其可能机制。方法:制备裸鼠移植瘤模型,随机分为生理盐水、氨基酸混合物、环磷酰胺、酪丝亮肽高低剂量等5组,每组12只,观察酪丝亮肽对人肝癌BEL-7402肿瘤组织坏死和凋亡的影响;电镜观察其对肿瘤细胞超微结构改变;流式细胞技术分析其对细胞周期及细胞凋亡率的影响。结果:酪丝亮肽能够诱导肿瘤细胞坏死和凋亡、细胞周期阻抑、显著抑制肿瘤的生长,给药剂量为80μg.kg-1.d-1和160μg.kg-1.d-1时,肿瘤抑制率分别为42%与58%。结论:酪丝亮肽可以显著抑制人肝癌BEL-7402的生长,作用机制与促进肿瘤细胞的凋亡和坏死有关。
Objective: To observe the inhibitory effect of casein leupeptin on human hepatocellular carcinoma BEL-7402 and to analyze its possible mechanism. Methods: Nude mouse xenograft models were established and randomly divided into 5 groups: normal saline, amino acid mixture, cyclophosphamide, tyroserleutifolium high and low dose groups, 12 in each group. The effects of tyroserleutide on human hepatocellular carcinoma BEL-7402 tumor tissue Necrosis and apoptosis. The ultrastructural changes of tumor cells were observed under electron microscope. The effect of apoptosis on cell cycle and apoptosis rate was analyzed by flow cytometry. Results: Tyroserleutide could induce necrosis and apoptosis of tumor cells, inhibit cell cycle and significantly inhibit the growth of tumor. When administered at doses of 80μg.kg-1.d-1 and 160μg.kg-1.d-1 , Tumor suppression rates were 42% and 58%. Conclusion: Tyroserleutide can significantly inhibit the growth of human hepatocellular carcinoma BEL-7402, the mechanism of which is related to the promotion of apoptosis and necrosis of tumor cells.
