目的探讨肾乐胶囊对慢性肾炎兔及慢性肾功能衰竭(肾衰)大鼠的治疗作用。方法利用兔慢性血清病肾炎模型、大鼠5/6肾切除肾衰模型、大鼠血瘀模型及小鼠水负荷模型,每个模型实验分为6组:正常对照组(假手术组),模型对照组,药物对照组,肾乐高、中、低剂量组。测定慢性血清病肾炎模型兔肾功能及24 h尿蛋白定量,ELISA法测定血清循环免疫复合物(CIC),MTT掺入法测定脾细胞白细胞介素2(IL-2)活性。测定5/6肾切除肾衰模型大鼠肾功能及24 h尿蛋白定量,并观察其肾脏病理改变。测定血瘀模型大鼠血液流变学指标及水负荷小鼠排尿量。结果与模型对照组比较,肾乐各剂量组给药6 wk血肌酐、尿素氮,24 h尿蛋白量降低(P<0.05),肾脏病理损害减轻,血清(白)蛋白升高(P<0.01),CIC和脾细胞IL-2活性下降(P<0.05)。肾乐各剂量组大鼠血液黏滞度均显著降低(P<0.01),水负荷小鼠给药后排尿量增加。结论肾乐胶囊能减轻肾小球炎症损害,改善肾功能,对慢性肾炎和慢性肾衰有一定疗效。
Objective To investigate the therapeutic effect of Shenle capsule on chronic nephritis rabbits and chronic renal failure (renal failure) rats. Methods The models of chronic serum nephritis in rabbits, 5/6 nephrectomy renal failure model, blood stasis model in rats and water stress model in mice were divided into 6 groups: normal control group (sham operation group) Model control group, drug control group, renal Lok high, medium and low dose group. Serum circulating immune complex (CIC) was measured by ELISA and the activity of interleukin 2 (IL-2) by MTT assay. The renal function and proteinuria of 24 h urinary protein in 5/6 nephrectomized rats were measured, and the renal pathological changes were observed. The blood rheology index and blood load in mice with blood stasis were measured. Results Compared with the model control group, the serum creatinine, blood urea nitrogen and 24 h urinary protein were decreased (P <0.05), renal pathological lesion was alleviated and serum ), IL-2 activity in CIC and spleen cells decreased (P <0.05). Renal music dose group rats were significantly lower blood viscosity (P <0.01), water-loaded mice increased urine output after administration. Conclusion Shenle Capsule can reduce glomerular inflammatory damage, improve renal function, chronic nephritis and chronic renal failure have a certain effect.
