目的对慢性丙型肝炎 (丙肝 )患者外周血单个核细胞 (PBMC)内病毒抗原进行鉴定及定位研究。方法应用抗 -HCV核心区和NS3单抗及光镜、电镜免疫组化技术对丙肝患者PBMC进行免疫酶标记。结果光镜免疫酶染色 ,HCVAg强阳性的PBMC内常规电镜首次发现其胞质空泡中直径 5 0~ 80nm具有双层膜的球型HCV颗粒和正在芽生病毒颗粒 ,均被免疫电镜酶反映产物特异性标记。结论免疫电镜证实 ,常规电镜所见病毒颗粒确为丙型肝炎病毒。从而为HCV感染PBMC并在其中复制提供了确切的形态学证据。
Objective To identify and locate viral antigens in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C (HCV). Methods Anti-HCV core region and NS3 monoclonal antibody and light microscopy, electron microscopy immunohistochemistry of hepatitis C in patients with PBMC immunoenzyme labeling. Results For the first time, it was found by immunoelectron microscopy that the globular HCV particles with diameter of 50 ~ 80 nm in diameter and budding virus particles in the cytoplasmic vacuoles were found by conventional electron microscopy in HCVAg strongly positive PBMC. Specific marker. Conclusions Immunoelectron microscopy confirmed that the virus particles seen by conventional electron microscopy were indeed Hepatitis C virus. Thus providing exact morphological evidence for HCV infection and replication in PBMCs.
