目的了解5岁以下结核病患儿血清中白细胞介素-35(IL-35)的表达和变化。方法用酶联免疫吸附试验检测56例结核病患儿及25例健康儿童血清样本中IL-35,根据光密度(OD)值计算相应IL-35浓度,分析各组间IL-35的差异。结果结核病组IL-35浓度为(505.52±660.77)pg/m L,健康对照组为(861.36±907.14)pg/m L,结核病组明显低于对照组(P<0.05);在56例结核病患儿中,肺结核35例,肺外结核21例,肺结核组IL-35浓度显著低于对照组(P<0.05),肺结核组IL-35浓度也低于肺外结核组,但差异无统计学意义(P>0.05);肺外结核组与对照组相比,IL-35的表达也发生了下降,但差异并无统计学意义(P>0.05)。结论 IL-35在5岁以下活动性结核患儿中的表达明显降低,且肺结核比肺外结核更显著,提示IL-35可能在结核病的发生和发展中扮演重要角色,上调其表达水平可能延缓结核病的进展,这为结核病的治疗提供了一条新的思路。
Objective To investigate the expression and changes of interleukin-35 (IL-35) in children under 5 years old with tuberculosis. Methods Serum samples of 56 children with tuberculosis and 25 healthy children were tested for IL-35 by enzyme linked immunosorbent assay (ELISA). The concentration of IL-35 was calculated according to the optical density (OD) value. The difference of IL-35 between groups was analyzed. Results The level of IL-35 in tuberculosis patients was (505.52 ± 660.77) pg / m L, compared with 861.36 ± 907.14 pg / m L in healthy controls and significantly lower in tuberculosis patients than in controls (P <0.05) Among the children, 35 cases were pulmonary tuberculosis and 21 cases were extrapulmonary tuberculosis. The concentration of IL-35 in tuberculosis group was significantly lower than that in control group (P <0.05), and the concentration of IL-35 in tuberculosis group was also lower than that in extra-pulmonary tuberculosis group, but the difference was not statistically significant (P> 0.05). Compared with the control group, the expression of IL-35 in extra-pulmonary tuberculosis group also decreased, but the difference was not statistically significant (P> 0.05). Conclusions The expression of IL-35 in children with active tuberculosis under 5 years old is significantly lower than that in children with pulmonary tuberculosis, suggesting that IL-35 may play an important role in the pathogenesis and development of tuberculosis.
