采用50个随机引物对奥利亚罗非鱼(Oreochromis aureus)和3个尼罗罗非鱼(Oreochromis niloticus)群体即无锡品系、埃及品系和吉富品系进行RAPD分析,共筛选出13个重复性好的引物,其中引物S1255扩增的2500bp片段具有群体特异性,可作为区分奥利亚罗非鱼和尼罗罗非鱼的分子标记;用PopGen1.32软件进行遗传多样性分析,结果表明:奥利亚罗非鱼、无锡品系、埃及品系和吉富品系罗非鱼群体内的多态位点率分别为22.03%、52.54%、58.47%、62.71%,基因多样性指数分别为0.0912、0.2166、0.2448、0.2705,Shannon信息指数分别为0.1323、0.3136、0.3537、0.3881。无锡品系、埃及品系和吉富品系罗非鱼群体内遗传相似性指数分别为0.8194、0.7886和0.7704,遗传变异水平较高,遗传多样性丰富。奥利亚罗非鱼的群体内遗传相似性指数为0.9279,群体的遗传纯度高,遗传变异水平较低。无锡、埃及、吉富品系尼罗罗非鱼与奥利亚罗非鱼之间的遗传距离分别为0.1370、0.2235,0.1986,说明奥利亚罗非鱼和埃及品系尼罗罗非鱼杂交可能产生更强的杂种优势。
Fifty random primers were used for RAPD analysis of Oreochromis aureus and three populations of Oreochromis niloticus, that is, Wuxi, Egypt and Jifu. A total of 13 reproducible . The 2500bp fragment amplified by primer S1255 was group-specific and could be used as a molecular marker for distinguishing O. tilapia from P. niloticus. PopGen1.32 software was used to analyze the genetic diversity. The results showed that: The polymorphic loci of Tilapia tilapia, Wuxi line, Egyptian line and Grifola line were 22.03%, 52.54%, 58.47% and 62.71% respectively, and the genetic diversity indices were 0.0912, 0.2166 and 0.2448 , 0.2705 and Shannon’s Information Index are 0.1323, 0.3136, 0.3537 and 0.3881 respectively. The genetic similarities index of the O. chinensis family, the Egyptian strain and the GIFT strain were 0.8194, 0.7886 and 0.7704, respectively. The genetic variation was high and the genetic diversity was abundant. The genetic similarity index of O. tilapia population was 0.9279. The genetic purity of the population was high and the genetic variation level was low. The genetic distances between Nile tilapia and Oreochromis niloticus in Wuxi, Gyeongdeok and Fujiwara strains were 0.1370, 0.2235 and 0.986, respectively, which indicated that the hybridization between O. tilapia and Egyptian strain Nile tilapia might be more Strong heterosis.
