本文对3种不同来源样品的4株椰酵假单胞菌的16SrDNA的序列进行了测定,以确定其系统发育位置及与其它菌种的遗传进化关系。实验中设计了7条引物,利用聚合酶链式反应(PolymaseChainRe-action,PCR)扩增4株细菌的16SrDNA,通过扩增产物的克隆测序或直接测序,获得了它们的16SrDNA序列,将测得的序列与伯克霍尔德氏菌属中其它菌种的16SrDNA序列进行了聚类分析,得到了系统发育进化树状图。树状图及不同菌种之间的同源性比值:椰酵假单胞菌(P.cocovenenanssubsp.farinofermentans)与唐菖蒲伯克霍尔德氏菌(Burkholderiagladioli)、椰毒伯克霍尔德氏菌(Burkholderiacocovenenans)的亲缘关系非常近,与其它菌种的亲缘关系相对较远,椰酵假单胞菌、唐菖蒲伯克霍尔德氏菌和椰毒伯克霍尔德氏菌可归属于一个独立的系统发育系。
In this paper, the sequence of 16S rDNA of four strains of P. aeruginosa from three different samples was determined to determine its phylogenetic position and its genetic evolution with other species. Seven primers were designed and used to amplify the 16SrDNA of four strains of bacteria by Polymerase chain reaction (PCR). The 16S rDNA sequences of the 16S rDNA sequences were obtained by cloning and sequencing of the amplified products. The phylogenetic tree was obtained by cluster analysis of 16SrDNA sequences of other species in Burkholderia. Dendrogram and homology ratio among different species: P.cocovenenanssubsp.farinofermentans and Burkholderiagladioli, Cotyledon Burkholde Burkholderiacocovenenans are very close relatives and are relatively distant from other species. Pseudomonas aeruginosa, Burkholderia gladius and Burkholderia coconut are attributed to An independent phylogenetic line.
