目的:研究鼠尾胶原对Caco-2细胞模型建立的影响,探讨适宜于Caco-2细胞模型建立的鼠尾胶原浓度。方法:将培养皿铺以750,250,50μg/ml三个浓度鼠尾胶原,各组接种相同数量的Caco-2细胞,培养21天,观察各组的细胞形态、跨膜电阻值和荧光素钠透过率的差异。结果:各组细胞在形态上没有差异,低浓度的鼠尾胶原能提高Caco-2细胞单层的跨膜电阻值,降低荧光素钠透过率(P<0.05)。结论:低浓度的鼠尾胶原可以增加Caco-2细胞的贴壁性,提高Caco-2细胞模型建立的成功率。
Objective: To study the effect of murine collagen on the establishment of Caco-2 cell model and to explore the concentration of murine collagen suitable for Caco-2 cell model. Methods: The culture dish was coated with three concentrations of rat tail collagen at 750, 250 and 50μg / ml. The same number of Caco-2 cells were inoculated in each group and cultured for 21 days. The cell morphology, transmembrane resistance and sodium fluorescein permeation Difference in rates. Results: There was no difference in morphology between the two groups. Low concentration of rat tail collagen increased transmembrane resistance of monolayer of Caco-2 cells and decreased sodium fluorescein transmittance (P <0.05). Conclusion: Low concentration of rat tail collagen can increase the adhesion of Caco-2 cells and increase the success rate of Caco-2 cell model establishment.
