目的探讨海参精囊水提物对模型小鼠睾丸生精细胞凋亡的影响及相关蛋白的表达情况。方法昆明种小鼠72只随机分为6组,除正常空白组用生理盐水外其他各组均腹腔注射环磷酰胺(CP,28mg/kg),每天1次,连续5 d,复制睾丸生精障碍小鼠模型;同时治疗组每天灌胃不同剂量的药物1次,连续4周。用TUNEL法检测各组小鼠睾丸生精细胞的凋亡情况;用免疫组化的方法检测Bcl-2和Bax的表达,并与模型组进行比较。结果海参精囊水提物能够抑制小鼠睾丸生精细胞的凋亡,使Bcl-2的表达增加,Bax的表达降低,与模型组比较有统计学意义(P<0.05、0.01)。结论海参精囊水提物对环磷酰胺所致生精障碍模型小鼠的生精细胞凋亡有一定的抑制作用,其作用可能与调控Bcl-2和Bax的表达有关。
Objective To investigate the effects of sea cucumber seminal vesicle aqueous extract on the apoptosis of spermatogenic cells in testes of mice and the expression of related proteins. Methods 72 Kunming mice were randomly divided into 6 groups. Except normal saline group, cyclosporine (CP, 28 mg / kg) was given intraperitoneally to all the other groups, once a day for 5 days, Obstructive mouse model; At the same time, the treatment group was given different dosages of drugs once daily for 4 weeks. The apoptosis of spermatogenic cells in testis was detected by TUNEL method. The expression of Bcl-2 and Bax was detected by immunohistochemistry and compared with the model group. Results The sea cucumber seminal vesicle aqueous extract could inhibit the apoptosis of spermatogenic cells in mice testes, increase the expression of Bcl-2 and decrease the expression of Bax, which was statistically significant compared with the model group (P <0.05, 0.01). Conclusion The water extract of sea cucumber seminal vesicle can inhibit the apoptosis of spermatogenic cells in cyclophosphamide-induced spermatogenic mice, which may be related to the regulation of the expression of Bcl-2 and Bax.
