使用荧光染料Indo-1测定法探讨了SiO_2对肺泡巨噬细胞胞浆游离钙水平的影响。结果表明,SiO_2对巨噬细胞胞浆游离钙水平的升高有明显的剂量效应关系。随着SiO_2作用时间的延长,胞浆游离钙水平呈持续性升高,并与培养液中LDH活性上升相平行。给予柠檬酸铝预处理的SiO_2可防止胞浆游离钙的增加。提示,SiO_2可引起巨噬细胞内钙稳态紊乱,这一紊乱与SiO_2对细胞膜的损伤有密切联系。
The effect of SiO_2 on the level of cytosolic free calcium in alveolar macrophages was investigated by fluorescent dye Indo-1 assay. The results showed that SiO 2 had a dose-dependent effect on the increase of cytosolic free calcium in macrophages. With the extension of the time of SiO 2, the level of cytoplasmic free calcium was continuously increased, and parallel with the rise of LDH activity in culture medium. Pretreatment of aluminum citrate with SiO 2 prevented an increase in cytoplasmic free calcium. Prompted, SiO 2 can cause macrophage calcium homeostasis, this disorder and SiO 2 on cell membrane damage are closely linked.
