Objective:Screening the different expression miRNAs as the biomarker for the diagnosisof salt-sensitive hypertension (SSH).Build a logistic regression model to predictthe SSH.Drawing the receiver operating characteristic curve to evaluate the accuracy ofthe model.Method: Modified acute oral NS-loaded test was used todistinguish the person of salt-sensitive and salt-resistant.First,samplesize of twelve underwent the miRNA sequence (Illumina Hiseq2000) and then used Quantitative Real-Time PCRto check the difference.SPSS21.0 was performed to run the two-independent t-test,Chi-square test and Binary Logistic regression.Finally,using the different expression miRNA as targets to draw ROC curves and cluster analysis by GraphPad Prism 6 and Cluster3.0.OmicsBean was the mean to predict the target gene along with pathway analysis.Results: There were 37 miRNAs that express differently between the two groups (SSHvs.SRH).8 miRNAs were eventually included (hsa-miR-15b-5p,hsa-miR-19a-3p,hsa-miR-210-3p,hsa-miR-26b-3p,hsa-miR-361-5p,hsa-miR-362-5p,hsa-miR-382-5p,hsa-miR-423-5p) to conduct the qRT-PCR.The expression quantity of hsa-miR-361-5p and hsa-miR-362-5p had significant differences between the two groups (P=0.037 and 0.049).Binary logistic regression panel:logit (P=SS) =1.67-2.395×hsa-miR-361-5p+0.948paste-0.707poultry.The odds ratio of hsa-miR-361-5p is 0.091,95% CI (0.014-0.595),P=0.012; The AUC of hsa-miR-361-5p was 0.793 when involving the paste and poultry (95% CI,0.698 to 0.888; sensitivity=73.9%,specificity=74.4%,P<0.001).The AUC of the 8 miRNAs was 0.836 when involving the frequency of having paste and poultry (95% CI,0.749-0.922; sensitivity=80.4%,specificity=81.4%,P<0.001).Conclusion:Has-miR-361-5p couldbe the most valuable biomarkerrelated to SSH.Although the prediction performance of hsa-miR-361-5p wasinferior to the 8 miRNAs,it was also a beneficial choice to predict SSH when considering its feasibility and economic effect.
