Platycodin D (PD) is the main saponin isolated from the traditional Chinese herb Platycodon radix.It exhibits anticancer activity in several types of cancer cell lines.Present study revealed that PD not only inhibited the cell proliferation in vitro but also retarded the growth of xenografts of hepatocellular carcinoma Bel-7402 cells.The increased apoptotic cells induced by PD were observed by using Annexin V/PI staining assay and Hoechst 33342 staining assay, respectively.The protein levels of cleaved PARP and cleaved caspase-3 were upregulated, while that of Bcl-2 was decreased after PD treatment.Moreover, PD triggered autophagy as evidenced by the formation of autophagic vacuoles, upregulation of microtubule-associated protein 1 light chain 3 Ⅱ, and the elevation of MDC-positive cells.Besides, PD-induced cell death was enhanced when combined with autophagy inhibitor, chloroquine or bafilomycin A1.These findings suggest that the combination with autophagy inhibitor and PD might be a promising strategy for the treatment of hepatocellular carcinoma cells.
