大鼠退变性髓核突出诱致神经根性疼痛及其机制研究

  目的 探讨大鼠正常与退变性髓核突出诱致神经根性疼痛的不同,研究椎间盘退变与神经根性疼痛之间的关系;探讨肿瘤坏死因子-α(TNF-α)及其受体(TNFR 1及TNFR 2)在疼痛进展过程中的作用。方法 利用尾椎椎间盘纤维环穿刺的方法建立椎间盘退变模型。分别取出自体的退变髓核与正常髓核组织,置于手术显露后的腰5左侧神经根处,建立髓核突出导致神经根性疼痛模型。分别在术后1,4,7,10,14和21天采用行为学测试的方法检测两组大鼠的运动功能、机械刺激阈值与热刺激阈值;分别在术后第4与14天采用免疫组化的方法检测两组的腰5左侧背根神经节中TNF-β的表达。
