高效液相色谱-电喷雾-四级杆-飞行时间串联质谱分析维药神香草化学成分

  目的:采用高效液相色谱-电喷雾-四极杆-飞行时间串联质谱法(LC-ESI-Q-TOF-MS/MS)对硬尖神香草乙醇提取物的主要化学成分进行分析鉴定.方法:加热回流法提取制备硬尖神香草乙醇提取物,Poroshell 120-C18 (2.7 mm×100rmm,2.7 μm)反相HPLC梯度洗脱法分离各主要成分;电喷雾电离源负离子模式对色谱流出物进行检测,四级杆飞行时间串联质谱法对各主要色谱峰进行归属.结果:通过一级和二级高分辨质谱分析结合对照品数据及相关文献,共鉴定出木犀草素、芹菜素、金合欢素-7-O-β-甲基葡萄糖醛酸苷等22个化合物.结论:LC-ESI-Q-TOF-MS/MS可从保留时间、精确相对分子质量、分子式和二级结构碎片等方面对硬尖神香草的主要成分进行定性分析,为寻找硬尖神香草中活性物质提供了一种快速准确的分析方法.
